Pro-inflammatory cytokine production by lip polysaccharide (LPS) in THP-1 cells was measured according to the method described by Hwang et al.27 During the assay, THP-1 cells were cultured in RPMI 1640 culture medium (Gibco BRL, Pasley, UK) containing 100 U/mL Penicillin and 100 mg/mL Streptomycin containing 10% fetal bovine serum (FBS, JRH). Cells were differentiated with phorbol myristate acetate (PMA, Sigma). Following cell plating, the test compounds 4-(4,6-dimethoxy-1,3,5-triazin-2-yl)-N-phenylpiperazine-1-carbox-amide urea derivatives (3a-h) and (4-(4,6-dimethoxy-1,3,5-triazin-2-yl)piperazin-1-yl)(phenyl)methanone amide derivatives (3i-p) in 0.5% DMSO were added to each well separately and the plate was incubated for 30 min at 37°C. Finally, LPS (E. coli 0127:B8, Sigma Chemical Co., St. Louis, MO) was added, at a final concentration of 1 μg/mL in each well. Plates were further incubated at 37°C for 24 h in 5% CO2. After incubation, supernatants were harvested, and assayed for TNF-α and IL-6 by ELISA as described by the manufacturer (BD Biosciences). The % inhibitions are measured at 10 μM concentration. The Dexamethasone is used as standard drug. The results are tabulated in Tables 15.7 and 15.10.

Tetrakis (hydroxymethy1) phosphonim chloride (THPC) aqueous solution, Gold (III) chloride trihydrate (HAuCl4.3H2O, ≥ 49% Au basis), 3-aminopropyltriethoxysilane (APTES), and Doxorubicin hydrochloride (DOX) are purchased from Sigma-Aldrich (USA). Dipalmitoylphosphatidylcholine (DPPC), cholesterol (Chol), and 1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[amino(polyethylene glycol)-2000] (ammonium salt) (DSPE-PEG2000) are purchased from Avanti Polar Lipids (USA). MCF-7 cell line is purchased from the Pasteur Institute (Iran). RPMI 1640 medium and fetal bovine serum (FBS) are purchased from GIBCO (USA). Annexin-V-FLUOS staining kit is purchased from Roche (Switzerland). Deionized water (18 MΩ.cm) is provided by a Milli-Q system and deoxygenated by vacuum for 1 hour prior to the use.

Hence, this method was additionally tested in exposure conditions very different from those described earlier. A volume of 0.45 mL of a mammalian cell culture medium (RPMI 1640 supplemented with 10% fetal bovine serum, obtained from American Type Tissue collection, Manassas, VA) was placed in an electroporation cuvette with 2-mm gap between two conductive walls (Bio-Rad Laboratories, Hercules, CA); the cuvette is shown in the inset of Figure 11.16. The conductive walls were connected to a Blumlein line pulse generator (custom built at the Old Dominion University, Norfolk, VA, see [41] for design details) to produce a high-voltage, 10-ns duration electrical pulse into the sample. The Blumlein line was charged from a high-voltage DC power supply (HR-100, American High Voltage, Elko, NV) until a breakdown voltage was reached across a spark gap in a pressurized switch chamber. The breakdown voltage (and, consequentially, the voltage of the 10-ns pulse delivered to the sample) was varied from 10 to 25–30 kV by increasing the pressure of SF6 gas in the switch chamber. The shape and voltage of the pulse delivered to the sample were monitored on a 500-MHz digital oscilloscope (TDS3052B, Tektronix, Wilsonville, OR) via a custom-made high-voltage probe. The pulse shape was close to rectangular, with some additional oscillations caused by impedance mismatch and the capacitance of the load.

Sorafenib (SAF) was purchased from Dalian Meilun Biotechnology Co., Ltd. (Dalian, China). Curcumin (CUR), Succinic anhydride, triethylamine (TEA), nile red and sodium dodecyl sulfate (SDS) were obtained from Aladdin Co., Ltd. (Shanghai, China). Poly (ethylene glycol) monomethyl ether (mPEG) and pyrene were provided by Jiuding Chemical Reagent Co., Ltd. (Shanghai, China). Polylactic Acid (PLA) was obtained from Pharmaceutical Sciences Institute Pilot Plant (Shandong, China). Glutathione (GSH) was purchased from Sigma-Aldrich (St. Louis, MO, USA). Dimethyl sulfoxide (DMSO) was purchased from Boster Biological Technology Co., Ltd. (Wuhan, China). Other reagents of HPLC grade were supplied by Tianjin Kemiou Chemical Reagent Co., Ltd. (Tianjin, China). Fetal bovine serum (FBS), penicillin-streptomycin solution, trypsin, Cell lysate (RIPA) were purchased from Beyotime Biotechnology Co., Ltd. (Shanghai, China). RPMI-1640 medium was purchased from GIBCO (Carlsbad, CA). BCA protein assay kit was purchased from ComWin Biotech Co., Ltd. (Beijing, China). Akt, p-Akt, and β-actin were purchased from Cell Signaling Technology (Beverly, MA). Horseradish peroxidase labeled goat anti-rabbit IgG was purchased from Zhongshan Jinqiao Biotechnology Co., Ltd. (Beijing, China).

Pluronic F127, F87, and P85 were obtained from the BASF Corporation (Parsippany, NJ, USA), for details about compositions, molecular weights (MW), hydrophilic–lipophilic balances (HLB), and critical micelle concentration (CMC) (see Table 1)[22,23] Phospholipid S100 was provided by Lipoid GmbH (Ludwigshafen, Germany). The human breast cancer cell line MCF-7 was obtained from Wuhan Procell Life Technology Co., Ltd. (Wuhan, China). RPMI-1640 medium was purchased from Gibco. Fetal bovine serum (FBS) was a product of Hangzhou Sijiqing Biological Engineering Materials Co., Ltd. (Hangzhou, China). Penicillin (100 U/mL) and streptomycin (100 µg/mL) were supplied by Hyclone (Logan, Utah, USA). MTT was purchased from Solarbio. Ethanol, cholesterol, and other reagent chemicals were all of analytical grade.
