Glut2

Glut2 is a protein that belongs to a family of proteins that help glucose enter pancreatic β-cells. It is a glucose transporter located at the plasma membrane of pancreatic β-cells and has a lower affinity for glucose, around 15-20mM, with symmetric kinetics. The insulin release from beta cells is triggered by the increased blood glucose concentration (Fig. 16.1 b). Glucose enters the beta cells through the glucose transporter 2 (GLUT2). As a substrate for glycolysis, glucose is quickly metabolized to generate high-energy ATP molecules, leading to a rise of the ATP/ADP ratio in the cell (Sweet et al., 2004). The increased intracellular ATP/ADP ratio causes the shutdown of ATP-gated potassium (KATP) channels in the cellular membrane, which prevents potassium ions (K+) from leaving the cell, leading to a buildup of potassium ions. The ensuring rise of positive charges inside cells leads to the depolarization of the cell surface membrane, which triggers the activation of voltage-gated calcium (CaV) channels. The brisk influx of calcium ion leads to the exocytotic release of insulin from their storage granule (Rajan et al., 1990).

GLUT2 is a glucose transporter located at the plasma membrane of pancreatic β cells. We analyzed the effect of TCDD on the level of glucose transporter 2 (GLUT2) associated with pancreatic β-cell function. Results showed that the level of GLUT2 was significantly decreased after exposure to 10 and 100 nM TCDD (Figure 8A). Meanwhile, oleuropein up-regulated the GLUT2 level inhibited by TCDD in INS-1 cells (Figure 8B).
