Nutrient Media for Cultivation of Industrial Microorganisms and Generation of Microbial Products

Blood consists of about 82% water, 0.1% carbohydrate, 0.6% fat, 16.4% nitrogen, and 0.7% ash. It is a waste product in slaughterhouses. Drying is achieved by passing live steam through the blood until the temperature reaches about 100°C. This treatment sterilizes it and also causes it to clot. It is then drained, pressed to remove serum, further dried, and ground. The resulting blood-meal is chocolate-colored and contains about 80% protein and small amounts of ash and lipids. Where sufficient blood is available, blood meal could form an important source of proteins for industrial media.

Blood meal (purchased from Guizhou Gold Wanhe Agricultural Science and Technology co., LTD, China) was used to simulate proliferation of soil microorganisms to help remediate organic contaminated soil in field experiments. Celery seeds, Libanotis seseloides (Fisch. & C. A. Mey. ex Turcz.) Turcz. used in this study, were purchased from Cephagri Seed (Jiaxing) Company Limited. A mixture of 16 US EPA priority PAH stock standard solutions (Nap, naphthalene; Ace, acenapthene; Acy, acenapthylene; Flu, fluorene; Phe, phenanthrene; Ant, anthrancene; Flr, fluoranthene; Pyr, pyrene; Baa, benz[a]anthracene; Chr, chrysene; Bbf, benzo[b]fluoranthene; Bkf, benzo[k]fluoranthene; Bap, benzo[a]pyrene; Daha, dibenz[a,h]anthracene; Bgp, benzo[g;h;i]perylene; Inp, indeno[1,2,3-cd]pyrene), purchased from AccuStandard (USA), were used as standard solutions to measure PAH concentrations. Solvents (analytically pure and chromatographically pure) were purchased from Sinopharm Chemical Reagent Co., LTD (Shanghai, China). Accelerated solvent extraction (ASE300, Dionex) and high-performance liquid chromatograph (HPLC Agilent 1100) were used to extract and analyze PAHs.
