Evaluation of precorneal residence time of brimonidine tartrate nanoparticles loaded in situ gel using gamma scintigraphy

In vitro stability of radio-labeling was performed by mixing 100 μl of 99mTc-in situ gel with 900 μl of simulated tear fluid (pH 7.2) saline. The mixture was allowed to vortexing for proper mixing and kept at room temperature. All the radiolabeling activity was carried out in a hot lab under lead shielding. Small aliquots were withdrawn at different time intervals up to 24 h. In vitro stability of labeled formulation was evaluated by standard ITLC method by using acetone as a mobile phase. The radioactivity of samples was counted by a gamma counter.[31]
