The hematopoietic stem cell niche is the name given to the complex multicellular and molecular microenvironment within the bone marrow that enables HSPCs to perform their functions of progenitor cell generation and self-renewal (Wei and Frenette, 2018). Numerous cell types resident in the bone marrow are integral to development and maintenance of a niche suitable for HSPC function (Figure 18.1). These include mesenchymal stem progenitor cells (MSPCs), which are the dominant producers of factors that promote HSPC activity, such as CXCL12 and stem cell factor. Additionally, cells of the osteoblast lineage (cells responsible for bone formation), endothelial cells, and adrenergic neurons, as well as the progeny of the HSPCs themselves, have roles in this process (Wei and Frenette, 2018).

Evidence exists that osteoclasts display several other functions. For instance, they produce clastokines that control osteoblast during the bone remodeling cycle. Recent evidence shows that osteoclasts may directly regulate the hematopoietic stem cell niche (Charles and Aliprantis 2014). This furthermore supports the theory of the bone being an endocrine organ.
