Also known as the INTERHEMISPHERIC FISSURE, this is the long FISSURE that separates the two HEMISPHERES of the BRAIN is the longitudinal fissure. Towards the posterior pole of the brain, where the right OCCIPITAL LOBE is separated from the left, it may be known as the median longitudinal fissure.

On PND 100, to collect brains for immunocytochemistry, the rats were administered i.p., a combination of ketamine (100 mg/ml, Imalgene®, Merial) 80 mg/kg, and xylazine (20 mg/ml Rompún®, Bayer) 10 mg/kg to achieve deep terminal anesthesia. Once the rat was deeply anesthetized, we accessed the heart. First, we injected 0.1 ml of heparin (Hospira®) into the left ventricle to prevent clotting. We connected the left ventricle to a vascular perfusion pump (Ismatec corp. Spain), which infused saline solution (0.9%) for 5 minutes, and then paraformaldehyde 4% in phosphate buffered saline (PBS) pH 7.4, for 20 minutes. Finally, brains were extracted and sectioned along their longitudinal fissure.

Animals were sacrificed by cervical dislocation, and the total rat brain was removed for isolation of hippocampus tissue as described previously [12]. Immediately, it was cut along the longitudinal fissure to divide both hemispheres on an ice-chilled plate. The exposed hippocampus tissue was resected by using scissors and microsurgical forceps from the neocortex. The whole hippocampus was homogenized in a 20-fold volume of formic acid (0.1 M) and homogenates were centrifuged at 18,000 × g for 20 min at 4 °C. The supernatants were collected and stored at −80 °C until analysis.

Uni- and bilaterality. CSD does not cross the medial longitudinal fissure. It remains in the hemisphere in which it is evoked [42]. However, migraine pain is unilateral in 60% of patients and bilateral in 40% of patients [86], indicating that it is not caused by putative CSD alone.
