Picoplatin is a cisplatin analog developed by Poniard Pharmaceuticals (previously NeoRx). It contains a 2-methyl-pyridine moiety in place of one of the amino ligands of cisplatin, and its origin was based on the concept of overcoming platinum drug resistance and making oral dosing possible.

Several drugs, such as irinotecan, paclitaxel, docetaxel, vinorelbine, gemcitabine, pemetrexed, picoplatin, have been evaluated in the second-line setting, with modest activity [63]. Recently, several phase II trials investigated chemotherapeutic agents such as bendamustine [64] and temozolomide [65]. The latter seems to be useful especially in patients with brain metastases and O6-methylguanine-DNA methyltransferase (MGMT) methylated, but more trials are needed to investigate this further.
