The pH of rice beer is measured directly using a pH meter. Total acidity and volatile acidity are measured by titration-based methods, which are conducted against sodium hydroxide solution. Both the results are expressed as grams of tartaric acid and grams of acetic acid present per 100 mL of the sample, respectively. Phenolphthalein is used as an indicator [13].

Persistent diarrhea may require evaluation for laxative abuse, not only in eating disorders but in any patient with unexplained diarrhea. Stools can be tested for phenolphthalein by alkalization using one drop of IN sodium hydroxide (NaOH) with 3 ml of stool supernate, which results in a positive pink to red color if present The presence of melanosis coli on sigmoidoscopy suggests prolonged use of laxatives. Of note, melanosis coli can occur in the sigmoid from anthracin (senna, cascara, and aloe), but not with phenolphthalein or bisacodyl, which are diphenolic laxatives (40).

Aquaporins are mainly expressed in the intestinal canal (Sun et al. 2020), which mediate the passive transport of free water across biofilm, thereby maintaining the homeostasis of the intracellular and extracellular environment (Matsuzaki et al. 2004). Moreover, the abnormal expression of aquaporins in the gastrointestinal tract is related to the occurrence of some diseases, such as constipation, gastritis, diarrhoea and gastric cancer (Liu et al. 2020). AQP3 is an important aquaporin located in the colon and is permeable to water (Li and Wang 2017). Previously studies have revealed the relationship between AQP3 and constipation, particularly morphine-induced constipation (Kon et al. 2015), promotes AQP3 expression level in the colon and subsequently increases water absorption from the luminal side to vasculature, which dries and hardens stool (Ikarashi et al. 2016). In the present study, the expression level of AQP3 was detected and the results showed that phenolphthalein and BCE relieve the symptoms of loperamide-induced constipation by decreasing the level of AQP3 in the colon of mice. However, the precise mechanisms remain to be further elucidated.

Citric acid, potassium sulphate, copper sulphate, sulphuric acid, bromocresol green, methyl red, ortho phosphoric acid, diphenyl amine indicator, ferrous ammonium sulphate, ninhydrin, stannous chloride, sodium citrate, n-propanol, potassium hydroxide, hexane, toluene, ammonium oxalate, potassium permanganate, ammonium molybdate, potassium nitrate, Folin-Ciocalteu's phenol reagent, sodium carbonate, ammonium acetate, diethylenetriaminepentaacetic acid (DTPA), calcium chloride, triethanolamine, and sodium azide were purchased from Merck India. Acetic acid, hydrochloric acid, sodium hydroxide, boric acid, petroleum ether, acetone, sodium sulphate, ammonia solution nitric acid, chloroform, and ethyl acetate were from Rankem, India. Potassium dichromate, methoxy ethanol, methanol, diethyl ether, and ethanol were supplied by S D Fine-Chem Limited, India. Dimethyl sulfoxide and phenolphthalein were procured from Ranbaxy, India. Leucine, pepsin, trypsin, and pancreatin were obtained from Himedia, India, and gallic acid was from Sigma Chemicals Co. (St. Louis, MO, USA).

The gastric content obtained from dissected stomach was centrifuged at 2500 rpm for 10 min to remove solid material. The pH of the supernatant of gastric juice was measured with help of pH meter. For the measurement of free acidity of gastric juice, 2–3 drops of methyl orange reagent was added to the gastric juice and titrated with 0.01 N NaOH till the appearance of pale color. The volume of added NaOH was noted. Then, 2–3 drops of phenolphthalein were added to the same solution for measurement of total acidity. The titration was carried out until pink color appeared. The total volume of NaOH added to the solution was noted. Acidity was measured with the help of the following formula (Katary and Salahuddin 2017).
