Spontaneous regression has also been observed in certain experimental tumors, like the malignant lymphomas induced in Swiss and NZW mice by 1-ethyl-1-nitrosourea (ENU) and the leukemia induced in chickens with avian myeloblastosis virus (AMV).285,286 The mechanisms involved in spontaneous regression of tumors are multiple and may be different in different cases but one mechanism may consist in the induction of terminal differentiation by unidentified endogenous and/or exogenous environmental stimuli. It is likely that only the tumors originated in partially differentiated cells can undergo spontaneous regression due to terminal differentiation.

The c-myb proto-oncogene is the cellular homologue of the v-myb gene of avian myeloblastosis virus (AMV). The v-myb oncogene transforms myeloid cells in culture and causes myelomonocytic leukemia in chickens; v-myb is truncated at its 5′ and 3′ ends relative to c-myb. The c-myb protein includes a conserved DNA binding domain at its 5′ end and a downstream “leucine zipper” motif, which is thought to mediate protein-protein interactions.74

The expression of ETA and ETB receptors mRNA was analyzed by RT-PCR. The expression of glyceraldehyde-3-phosphate dehydrogenase (GAPDH) mRNA was determined as an internal control. Total tissue RNA was isolated by acid guanidinium thiocyanate-phenol-chloroform extraction with Isogen according to the manufacturer’s instructions. The tissue was homogenized in Trizol (0.2 g tissue per 2 ml Trizol) with a tissue homogenizer. Chloroform extraction, isopropanol precipitation, and 75% (v/v) ethanol washing of precipitated RNA were subsequently performed. The obtained RNA was resolved in diethyl pyrocarbonate–treated water. The RNA concentration was measured using nanodrop. Total RNA (5 µg) was primed with 0.05 µg oligo-d(T) 12–18 and reverse transcribed by avian myeloblastosis virus reverse transcriptase using the First-Strand cDNA Synthesis Kit. The obtained cDNA was diluted in a 1:10 ratio, and 1 µl was used for real-time PCR.

Total RNA of cells was extracted using Trizol (Invitrogen). Ultraviolet analysis and formaldehyde denaturing gel electrophoresis were applied to determine the high quality of obtained RNA. Next, l μg RNA was reverse transcribed into cDNA using avian myeloblastosis virus reverse transcriptase. The SYBR Green method was used for qPCR. PCR primers (Table 1) were synthesised by Sangon (Shanghai, China). Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) and U6 were set as the internal reference. Data were analysed using 2−ΔΔCt method in which ΔΔCt = [Ct (target gene) − Ct (internal control)]experimental group − [Ct (target gene) − Ct (internal control)]control group.
