In interpreting such morphometric data related to secretory granules and products of submucosal gland, it should be noted that the following possibilities may account for the differences. First, there is more connective tissue or inflammatory or other nonsecretory cells in the more proximal human glands or in the female rat glands, so the proximal or female secretory cells as a whole must have contained relatively less acid mucin and relatively more cytoplasm. Second, the individual secretory granules in the mucous cell will vary in their rates of production and excretion and their initial volumes. For example, Phipps et al. (1986) have studied effects of O3 exposure on sheep tracheal mucus secretion by both measuring radiolabeled glycoprotein release and morphometry of submucosal glands. Although chronic exposure produced glandular hypertrophy, it also induced a decrease in glycoprotein release and the gland cells were devoid of secretory material (granules). With recovery, basal glycoprotein secretion was greatly increased above control and gland cells were full of secretory material.

The histopathological features studied include the quantity of endometriotic foci, components, phasing of the endometriotic foci, and stromal features.Quantity of endometriotic foci (severity): Assessed on the entire tissue examined and graded based on the extent occupied in low power field (LP) as (i) mild (one LP field), (ii) moderate (2–3 LP fields) and (iii) severe (>3 LP fields).The components in the endometriotic foci: classified as (i) pure glandular (when only glands were present without stroma), (ii) pure stromal (only stroma was present without glands), and (iii) mixed stromal and glandular (both gland and stroma were present). The type of gland seen was also classified as endometrial type (well-differentiated glands), undifferentiated type (when glandular component resembled other mesothelial types), and glandular pattern of mixed differentiation (both endometrial and undifferentiated glands present). The presence and absence of haemorrhage and hemosiderin was recorded.Phasing of the endometriotic foci: inactive, proliferative, or secretory, similar to that of eutopic endometrium. The presence of atypia was noted.Stromal features: Presence of oedema, fibrosis, and inflammation was noted.

Morphological and immunohistochemical examinations were performed in the Department of Pathology, Faculty of Medicine, Adıyaman University. Tissues were initially fixed with 10% formalin. Each sample block was then dehydrated with a series of increasing alcohol concentrations. Xylene was then used to clean the sections, and the blocks were embedded in paraffin. A Thermo Shandon Finesse ME microtome (Thermo Fisher Scientific, Cheshire, UK) was used to cut sections from the blocks with a thickness of 3 μm. Sections were stained with haematoxylin-eosin and examined under a light microscope by a pathologist (BAT) who was blind to the randomisation of the groups. For microscopic examinations, an Olympus BX53 microscope (Olympus Corporation, Hamburg, Germany) was used. Glandular density, the presence of endometrial hyperplasia, the presence of squamous metaplasia, the presence of luminal stratification, epithelial neutrophil infiltration, and the number of epithelial mitosis (x40 magnification area) were evaluated in microscopic examinations of the endometrium. Glandular density was defined as the number of glands where it was most intense in the area of magnification (x40) and the presence of luminal stratification was defined as an increase in the lining of the epithelium covering the uterine cavity.

The experimental and control groups are outlined in Table 1. As shown in Figure 1A, a trimmed segment of endometrial tissue was sutured to the inner side of the abdominal wall. We noted that an ectopic mass in the abdominal wall adhered to the surrounding connective tissues, and neovascularization developed in the surface of the mass (Figure 1B, C), which showed that a model of endometriosis was established. The cyst was 5–10 mm in diameter and contained a clear or purplish-red fluid. Figure 1D shows the vaginal plug, which is an important marker of the endometrium entering the window of implantation (WOI). Hematoxylin-eosin staining analysis, showed the growth of glands in the ectopic mass (Figure 1E, F), modeling endometriosis. The glandular epithelial cells were columnar or cubic, and inflammatory cells and secretions existed in some glandular lumen.
