Methods have been developed to purify DNA from organisms, such as phenol-chloroform extraction, and to manipulate it in the laboratory, such as restriction digests and PCR. Modern biology and biochemistry make intensive use of these techniques in recombinant DNA technology. Recombinant DNA is a man-made DNA sequence that has been assembled from other DNA sequences. They can be transformed into organisms in the form of plasmids or in the appropriate format, by using a viral vector. The genetically modified organisms produced can be used to cultivate in agriculture or to produce products such as recombinant proteins that are used in medical research (Figure 2.13).

For molecular analysis, Cryptosporidium DNA has been extracted using various methodologies. Hemolymph or gill washings were subjected to five freeze-thaw cycles, followed by phenol-chloroform extraction. The extracted DNA was evaluated using small subunit (SSU) rRNA-based PCR restriction fragment length polymorphism (RFLP). DNA also has been extracted using other DNA extraction kits; among them are the QIAamp DNA minikit (Qiagen Valencia, CA) or FASTDNA Spin KIT for Soil (MP Biomedicals, Irvine, CA) (Jiang et al., 2005).

Bacterial cells were harvested by centrifugation from the cultures at each sampling time. DNA was extracted from the pelleted cells using phenol-chloroform extraction.[7,23] Total DNA was also extracted from the Pdy soil (250 mg) using the Powersoil Extraction Kit (MoBio) following the manufacturer’s instruction.[27,28]
