Human breast and prostate adenocarcinoma cells— The human breast adenocarcinoma cell line MCF-7, passages 6 to 14 and 78 to 88, and HBL-100 were obtained from Dr. David Kingsbury at Naval Biosciences Laboratory, Oakland, and from the Mason Research Institute, Worcester, Mass. Prostate carcinoma cell line PC-3 was established from a poorly differentiated human prostate adenocarcinoma which had metastasized to bone.14

PC-3 cells are highly invasive and metastatic with a mesenchymal phenotype that is appropriate for anticancer agents (Tai et al. 2011). Our previous study showed that the treatment of PC-3 cells with methanolic extract of T. persicum leads to membrane localization of β-catenin protein and induction of epithelial-like morphology (Barati et al. 2021). Regarding the β-catenin as a partner of E-cadherin protein in adherence junctions (Zhu et al. 2020), we decided to evaluate the effect of T. persicum methanolic extract on the expression and cellular localization of E-cadherin protein in PC-3 cells.

There are some PCa cell lines that are used for research purposes. PC3 with a doubling time of approximately 33 h[21] is a cell line isolated from a vertebral metastatic prostate tumor in 1979.[22] It is also hormone insensitive and presents no androgen receptors (AR) or PSA mRNA/protein. It was first described as a human prostatic adenocarcinoma metastatic cell which has a greatly reduced dependence upon serum for growth.[23]
