Baeyens et al. [172] reported that fluid shear stress potentiates BMPs to activate Alk1 signaling, with enhanced association of Alk1 and endoglin. Alk1 is required for BMP9 and flow responses, while endoglin is required for enhancement by flow [172]. Impaired Alk1 signaling results in the formation of vascular shunts, specifically in regions of high blood flow [172]. Overall, Baeyens et al. [172] reported that Alk1-endoglin complex has a joint function of BMP/flow receptor, regulating vascular morphogenesis through vessel stabilization. Further research on the mechanism through which flow acts on these receptors enhancing BMP signaling may identify new targets of therapy through the understanding of vascular mechanotransduction [172].
