Metagenomics was first introduced and practiced by Handelsman in 1998 in their report on biochemical diversity of soil microbes; however, earlier report on metagenomic was proposed by Pace et al. in 1985. They reported the metagenomic sequencing of ribosomal RNA with the existing 5S rRNA sequences. The phylogenetic affinities of organisms contributing to the analyzed 5S rRNAs were defined in the first approach. This was is followed by analyzing the mixed populations using 16S rRNA genes with shotgun-cloned using total DNA isolated from mixed microbial populations.
