Tooth Whitening Materials

Some products in this category contain titanium dioxide, a white pigment found in paint. It acts by entering the surface irregularities of the tooth (Haywood 1995b) and gives the illusion of whitened teeth. Only a surface phenomenon, titanium dioxide does not penetrate internally and thus does not modify the internal color or whiten teeth. Other ingredients include tetrasodium pyrophosphate for the soothing effect on the teeth as well as sodium lauryl sulphate for the foaming capability and hydrated silica.

Western blot analysis were performed using roughly 30 mg of the rear half of a mouse brain hemisphere. Brain homogenate was prepared in lysis buffer (1 mM EDTA, 0.5% Triton X-100, 5 mM NaF, 6 M urea, 2.5 mM tetrasodium pyrophosphate, 1 mM sodium orthovanadate, 0.5% sodium deoxycholate, 0.5% sodium dodecyl sulphate in phosphate-buffered saline) with freshly added protease inhibitors (aprotinin 1.7 mg/ml, leupeptin 5 mg/ml, pepstatin 5 mg/ml, PMSF 100 lM). Samples were electrophoresed on 12% Mini-PROTEAN® TGXTM precast gels (BioRad, Munich, Germany) after application of 10 µg protein per lane and transferred to polyvinylidene fluoride (PVDF) membranes (Roth, Karlsruhe, Germany). Afterwards, the membranes were incubated with the respective primary antibodies from Abcam, Cambridge, UK; (Catalase antibody (ab209211), GPX-1 antibody (ab108427), SOD2 antibody (ab13533)) and secondary antibodies (Calbiochem, Germany) conjugated to horseradish peroxidase. Antibodies were diluted according to manufacturer’s instructions and processed for visualization by Luminata reagents (Millipore, Darmstadt, Germany). Tubulin served as a loading control. Band analysis was performed using BioRad’s Quantity One software.

Cells were lysed in 20 mM HEPES, pH 7.5, 150 mM NaCl, 1% NP-40, 10 mM tetrasodium pyrophosphate, 100 mM NaF, 17.5 mM 1-glycerophosphate buffer supplemented with Complete Mini Protease Inhibitor Cocktail tablets (Roche Applied Sciences, Indianapolis, IN). Samples separated by SDS-PAGE were transferred to nitrocellulose membranes, blocked with 5% bovine serum albumin (w/v) at room temperature for 1 h, and incubated with primary antibodies (1:1000 dilution) at 4 °C overnight. After incubation with secondary antibody (1:3000 dilution) at room temperature for 1 h, the membranes were developed with chemiluminescence ECL reagent (Lumi Gold, Signa Gen Laboratories, Rockville, MD) and exposed to Hyper film MP (GE Healthcare Life Science, Pittsburgh, PA).

MY, MG, and SIII with a purity of >90% were purchased from Thermo Fisher Scientific (Mumbai, India). Ethylene glycol-O,-O′-bis(2-aminoethyl) N,N,N′,N-tetraacetic acid (EGTA) was purchased from Sisco Research Laboratory (Mumbai, India). All the routine laboratory chemicals including reduced nicotinamide adenine dinucleotide (NADH), 5,5-dithiobis (2-nitrobenzoic acid) (DTNB), sodium dodecyl sulfate (SDS), nitroblue tetrazolium chloride (NBT), phenazine methosulfate (PMS), tetrasodium pyrophosphate, benzylamine, cresyl violet, Triton-X 100, and Follin’s reagent were purchased from Sigma-Aldrich (St. Louis, MO) and CDH (New Delhi, India).
