Ferroptosis is a novel form of cell death identified in 2012, which differs from both apoptosis and necrosis with the characteristic of iron-induced lipid peroxidation [3]. The activation of ferroptosis has been demonstrated in some pathological states, thus its dysregulation is involved in a wide variety of pathological processes, such as lung injury, neurodegeneration, and renal failure [4,5]. Interestingly, tumor cells can accommodate the oxidative condition to restrict ferroptosis, which may remodel the tumor niche to promote cancer proliferation and development [6]. Therefore, agents that can activate ferroptosis in tumor cells have been verified to be the new antitumor therapeutic options. For instance, ketamine activates ferroptosis in BC that induces the inhibition of cell growth via regulating KAT5/GPX4 pathway [7]. The same inhibitory effect of 18-β-glycyrrhetinic acid [8] and simvastatin [9] on the proliferation of triple-negative breast cancer is confirmed by activating ferroptosis. The repressive role of different agents in the cell proliferation of different cancers through the activation of ferroptosis is also reported, such as tanshinone IIA in gastric cancer [10], curcumin in non-small-cell lung cancer [11], and auriculasin extracted from Flemingia philippinensis in colorectal cancer [12].

Genistein was first isolated from dyer’s broom Genista tinctoria plant. The main sources are soybeans, fava beans, and psoralen. Also founds in many medicinal plants like Flemingia vestita and Flemingia macrophylla (83). Genistein indicates chemo-therapeutic and chemo-preventive exercises in numerous tumors, particularly in hormone-dependent cancers. The in vivo and in vitro studies conformed its antineoplastic effects in prostate, breast, urinary, colon, and skin cancer (84). Tian et al. (85) reported that in lung cancer treatment genistein (25–75 μM) could inhibit the cell proliferation and migration of H446 cell by the induction of cycle arrest at G2/M-stages and apoptosis. In the prostate malignancy (LNCaP and PC-3) and esophageal squamous cell carcinoma (KYSE-150), genistein repress the DNMT by the reduction of cellular accessibility of SAM with the dose of 20–50 μmol/L (86). In the breast cancer the conjugates of EGF-genistein could inhibit the EGF-Rc tyrosine kinase and induce the apoptosis, killing approximately 99.99% of clonogenic breast cancer cells (87). A study revealed that in the treatment (25–100 μM) of colon cancer cells, genistein exhibit pro-apoptosis and anti-proliferative effects (88).

The results revealed the apoptotic nature of BH-AgNPs by fluorescent staining when compared with treated KB cells to untreated cells, they show more number of early apoptotic cells and late apoptotic cells and necrotic cells with chromatin condensation and blebbing of the cell membrane in the human oral carcinoma cells was very clearly observed (Figure 8(a). The final results were similar to that of standard drug Camptothecin treated cells indicating that BH-AgNPs have very good anticancer activity. Similarly, the biosynthesized AgNPs using leaf extract of Flemingia wightiana, Saccharina japonica and Asian Spider Flower showed apoptotic cells and late apoptotic cells with blebbing of the cell membrane, chromatin condensation, followed by necrotic cells [55–57].
