With regards to the research in the field of neglected disease, the simple derivative dimethylcurcumin (32) (Figure 9) resulted more potent than curcumin (1), by reducing of 66% the lesion size in Leishmania amazonensis infected mice at a subcutaneous dose of 20 mg/Kg/day (Araújo et al. 1999). The LD50 of 2.0 μg/mL is reported for this compound on promastigote forms of Leishmania amazonensis (Gomes et al. 2002). Additionally, the derivative (33) (i.e., 1E,4Z,6E)-5-hydroxy-1-(4-hydroxy-3-methoxyphenyl)-7-(4- methoxyphenyl)hepta-1,4,6-trien-3-one; Figure 9) was also tested in a model of Leishmania amazonensis, demonstrating high activity in vivo as anti-Leishmania agent (Alves et al. 2003, 2004). Curcumin was reported to have antiandrogenic activity [99,100], and several series of curcumin derivatives have been developed. AndroScience reported dimethylcurcumin (ASC-J9: 19) derivatives bearing substituents at the central carbon atom [101]. These compounds reduced the expression of AR protein in the prostate cancer cell lines LNCaP and 22Rv1 (Figure 23) [102].
