The nature of this phenomenon, which could provide novel approaches to understanding the well-known beneficial health effects of tea phenols, is investigated in this study, and it was found that selected polyphenols displayed affinity for all of the selected cell nuclear structures. Theaflavin-digallate was shown to display the highest affinity for DNA reported for any naturally occurring molecule reported so far. This finding may have implications for understanding the role of tea phenolics as “life span essentials” (Mikutis, Karaköse, Jaiswal et al. 2013a).

Shetty et al.6 reported that the green tea extracts inhibited the growths of diarrhea causing S. aureus, S. epidermidis, V. parahaemolyticus, C. jejuni and V. cholera. Antibacterial activity of tea polyphenols against C. botulinum was studied by Yokihiko and Watanabe.7 They found C. botulinum spores were killed when inoculated into tea drinks. Horiuchi et al2 examined the bactericidal activity of tea and catechins against B. pertussis. Green tea, black tea and coffee showed marked bactericidal activity at their concentrations in beverages, while puerh tea killed the bacteria in a moderate way. This study suggest that green tea, black tea, epigallocatechin gallate (EGCG) and theaflavin digallate (TF3) might act as prophylactic against pertussis infection.

Second messenger c-di-AMP and Ap4A regulate S. mutans biofilms formation. Inhibitors targeting synthetase/hydrolase or receptor proteins can affect the intracellular levels or signaling transduction of second messengers and inhibit biofilm formation. Currently, most c-di-AMP inhibitors target PDE and DisA [126]. For example, three inhibitors of Bacillus subtilis DisA, bromophenol-TH, suramin, and the tea polyphenol theaflavin digallate, were identified [126]. Stress alarmone ppGpp could competitively inhibit B. subtilis GdpP (PDE) [127]. However, inhibitors targeting S. mutans DAC/PDE still need further investigation. The mechanisms of action of these inhibitors and whether there might be an impact on the other flora in the oral cavity are still unclear.

In MD experiments, Mhatre et al. (2021) found that EGCG binds tightly to 3CLpro, PLpro, RdRp, spike RBD, and RBD-ACE2 of SARS-CoV-2. Compared with the RdRp inhibitors remdesivir and favipiravir, EGCG and theaflavin digallate have higher binding affinities for RdRp than remdesivir and favipiravir.
