Early afternoon on the Florida Turnpike, I drove the Audi on cruise control, seat belt fastened, my husband right behind me in the Mercedes. All of a sudden–a tremendous jolt! CRASH… BANG… I wasn’t sure what had happened. I was turning and tumbling, rising, falling–then came to a stop. I was conscious and aware of still being in the car, but felt upside down … I heard a voice: “Don’t move, we’ll get you out–you’re trapped in the car. You had an accident!” It was my husband trying to comfort me. I felt a stabbing pain in my neck, a roaring noise in my ears–panic. I suddenly recalled a similar sensation years ago when swimming in the ocean, not far from the shore. I got caught in an undertow, going down, down… What was happening now? … Dimly I heard other voices. A trucker had stopped and attempted to pull me out but couldn’t reach me. An emergency wrecker arrived and literally cut me out of the car with saws. The Audi was a total wreck. The roof was smashed in and broken glass everywhere. Yet I did not have one scratch–ONLY A BROKEN NECK! I remember repeating “Help me, help me, I’m in such pain.” I never lost consciousness, but things were blurry around me. After what seemed an eternity, an ambulance with sirens at full blast rushed me to the nearest hospital about 15 miles from the scene of the accident.
Neuroprotective effect of miR-212-5p on isoflurane-induced cognitive dysfunction by inhibiting neuroinflammation

Adult male Sprague-Dawley (SD) rats (7–8 weeks, weighted 210–250 g) were supplied by Sino-British Sippr/BK Lab Animal Ltd (Shanghai, China) and housed in an air-conditioned animal quarter at 20–24 °C with the relative humidity of 40–60%. During this period, animals had free access to food and water. Animals were divided into four groups randomly with 16 rats of each, including a Control group (air inhalation for 6 h), an isoflurane group (3% isoflurane inhalation for 6 h), an agomir NC group (50 μM miR-212-5p agomir negative control (RiboBio Co., China) injection with 3% isoflurane), and a miR-212-5p agomir group (50 μM miR-212-5p agomir (RiboBio Co., China) injection with 3% isoflurane) according to previous studies (Ma et al. 2017; Hu et al. 2018; Zhao et al. 2018; Si et al. 2020). The flow rate of isoflurane 2 L/min according to previous study with partial improvement (Li et al. 2015).In all rat models, the artery blood gases concentration, body temperature, and blood pressure were tightly monitored and maintained in the normal range during anesthesia. Isoflurane was purchased from Sigma-Aldrich Corp. (St. Louis, MO, USA.). Ten rats were selected from each group for following experiments. The rats were sacrificed by broken neck and the hippocampi tissues were dissected from the rats and placed on ice immediately for further analyses.

To assess whether SSFH/Cas9 could inhibit AML proliferation and prolong retention in vivo, 2 × 107in vitro-propagated Molm-13 cells were injected subcutaneously into the flank of BALB/c mice to generate the mouse xenograft tumor model. Fourteen days later, treatments were initiated. The mice were divided into four groups, with six in each group: ① treated with saline once a day; ② treated with SS30 every 3 days (2 mM/kg); ③ treated with SSFH every 3 days (2 mM/kg); ④treated with SSFH/Cas9/sgRNA once every 3 days (2 mM/kg). The agents were administered through tail vein injection. The tumor volume was calculated using the following equation: tumor volume = tumor length × tumor width2/2. The body weights and survival rate were assessed and calculated. Subsequently, when the tumor volume was over 2000 mm3, or the mice that lost over 15% of their pretreatment body weights, the mice were euthanized by broken neck. Tumor tissues were collected and subjected to JAK2/STAT5 staining to evaluate anti-cancer effects of SSFH.
