Top off the tube with HBSS and centrifuge at 250 × g for 5 min. Wash the cell pellet twice with HBSS and adjust the lymphocyte concentration for HLA typing. The authors have found that freshly made Tris ammonium chloride is not reliable and that "matured" Tris ammonium chloride (>3 months old) is far more efficient.

Information regarding the use of alkalinizers other than bicarbonate is scarce. Citrate has been used, with or without bicarbonate, and trihydroxymethylaminomethane (TRIS) has also been used, but it is a potent diuretic. Caution with ingestion of large doses of sodium bicarbonate is necessary. Excessive doses have produced nausea, vomiting, flatulence, diarrhea, and even muscle cramps.355,362 Also, determination of urine pH (a very simple task) is sufficient to detect athletes who use alkaline loading, although no sports governing agency yet considers this practice to be doping.

Phosphate buffered saline (PBS) of pH 7.4 and osmolality 290 to 300 mmol/kg is commonly used although the zwitterionic HEPES buffer has advantages.102 Tris buffer should be avoided since it causes a time- and concentration-dependent increase in erythrocyte volume. Once resuspended in buffer, erythrocyte deformability should be measured as soon as possible. It should also be remembered that washing erythrocytes in buffer may remove membrane lipid as well as adherent protein and this has been reported to impair filterability;104 increased resistance to osmotic stress has also been shown for washed erythrocytes.61,104

In this study, we used a higher concentration of Tris buffer (50 mM), compared to 10 mM Tris buffer used by Nilam et al.,27. To test the possibility that Tris is competing with other components for binding to CB6, we directly compared 10 mM Tris buffer and 50 mM Tris buffer in the traditional and the optimised ODC tandem assays. We found that the traditional assay showed DMSO-dependent fluorescence quenching regardless of Tris concentration (Figure 5A–D). In marked contrast, the optimised assay showed no DMSO sensitivity in both 10 mM and 50 mM Tris buffers (Figure 5E–H). Although DMSO sensitivity of the traditional assay was independent of Tris concentration, we found that 10 mM Tris buffer showed 1.5 to 2-fold larger dynamic range than 50 mM Tris buffer in both the traditional and the optimised assays (Figure 5A, C, E and G). These results indicate that Tris is likely to induce a mild fluorescence quenching, which should be taken into account in future optimizations of the assay.

Olfactory cilia isolation was performed using the Na+ and Ca2+ shock method as described by Kuhlmann et al. [19,20]. Briefly, OMs from 6 mice were pooled and washed with an ice-cold solution containing 20 mM HEPES, 140 mM NaCl, 3 mM KCl, 2 mM MgSO4, 7.5 mM glucose and 5 mM EGTA (solution A; pH 7.4). They were then resuspended in solution A supplemented with 20 mM CaCl2 and 30 mM KCl. Cilia were detached by gentle stirring for 20 min at 4 °C and isolated by centrifugation for 5 min (6,600 g, 4 °C). The supernatant was collected, and the deciliation procedure was repeated five times. The supernatants were pooled and centrifuged at 45,000 g for 30 min at 4 °C. The cilia were resuspended in 500 µl of Tris-EDTA buffer and used immediately or stored at −80 °C until further analysis.
