Polyhydroxybutyrate-Based Nanoparticles for Controlled Drug Delivery

Dipyridamole is used as a model drug for studying controlled release formulations with a PHB nanoparticle matrix. Dipyridamole (molecular formula: C24H40N8O4 with molar mass 504.625 KDa) is an odorless, yellow crystalline powder having a melting point in the range of 164–168°C and is soluble in dilute acids, methanol, ethanol, and chloroform. It is a phosphodiesterase inhibitor that blocks the uptake and metabolism of adenosine by erythrocytes and endothelial cells. Dipyridamole inhibits the formation of blood clots (Qiu et al. 2014). The drug can be administered in oral or intravenous form for the treatment of cardiovascular, hypertension, and stroke patients. At high doses over a short time, it inhibits the phosphodiesterase enzymes that normally break down cAMP by increasing cellular cAMP levels and blocks the platelet aggregation response to ADP and cGMP.

Bakola et al. fabricated a drug delivery nano-system consisting of electrospun hydrophobic polylactic acid (PLA) fibrous scaffold loaded with dipyridamole (a nucleoside transport inhibitor and a phosphodiesterase enzyme-3 (PDE3) inhibitor), added during the electrospinning process.32 The surface morphology and topography were found to be suitable for tissue engineering, and further studies revealed that the fibrous nanoplatforms enhance cytocompatibility when used with mice fibroblasts (L929) and are capable of sustaining a controlled release of dipyridamole.
