Adefovir

Adefovir is a medication used to treat Hepatitis B virus. It was previously known as bis-POM PMEA and is prepared with lipid nanoparticles. It is also referred to as Adefovir dipivoxil, which is an ANP-based medicine available as an ester prodrug for oral administration. Lipid nanoparticles are in solid-state at body temperature, used as a carrier for drug delivery (Figure 8.4). Lipid nanoparticles are more advantageous when compared to liposomes. Adefovir dipivoxil was previously known as bis-POM PMEA, prepared with lipid nanoparticles, and is used to treat the Hepatitis B virus [5].

Several ANP-based medicines are also available as ester prodrugs for oral administration, such as the bis(pivaloyloxymethyl)ester [–CH2–O–C(O)–C(CH3)3] of PMEA, also known as Adefovir dipivoxil (Hepsera) [15], which was approved for treatment of hepatitis B infection in 2002 (FDA) and 2003 (EMA) [21]. Likewise, the prodrug bis-(hexadecyloxypropyl)-Cidofovir (trade name Brincidofovir), also suitable for oral administration, is being developed against orthopoxvirus infections and has already been employed to combat Ebola virus infections during the 2014 epidemic [22].
