Autologous Stem Cell Transplantation in Animal Models of Autoimmune Diseases

The predictive value of experiments with animal disease models is generally felt to be greater if the causes and nature of the model disease have more resemblance to that of the human disease. Many of the human autoimmune diseases are T cell initiated or T cell mediated. In the majority, the presence of auto-antibodies in the serum is considered to be an associated- or epiphenomenon. The mechanisms of activation of T lymphocytes against self-antigens are largely unknown but certainly diverse. It may be induced by the release of an excessive amount of tissue specific antigens to which the organism has not become tolerant during development, for instance as in sympathetic ophthalmia.

In the context of sympathetic ophthalmia, recently Brar et al. have used widefield OCTA to assess treatment response following corticosteroids and immunosuppression. On sequential imaging, flow voids in the choriocapillaris improved following initiation of corticosteroid and immunosuppressive therapy. Complete resolution was seen following 6 months of therapy with a corresponding improvement in visual acuity. They suggested that ischemic changes in choriocapillaris can be used as anatomical markers to monitor treatment response [34]. Thus, SS-OCTA appears invaluable in sympathetic ophthalmia as well as in VKH disease.
