Semen collection is an essential part of preservation protocols and AI programs. The quality of collected semen depends on numerous factors such as libido, season, age, or breed (Samper, 2000). Semen can be collected using different methods; condom, pharmacological induction, manual manipulation or by the use of artificial vagina, the tool most commonly used (Samper, 2000). There are several models of artificial vagina available commercially. The Colorado model is the most widespread at the beginning, but currently the most used is the Missouri model due to the improvements and efficiency.
Practical problems: ‘how to do it’

For the vagina to be accessible, each partner needs to be able to get into a position such that penetration is possible (see Positions). If the difficulty is due to restricted movement because of pain (e.g. arthritis of the hip), adjusting the medication to gain maximum freedom from pain and maximum mobility can be helpful. If there is no accessible or available vagina, then the possibility of a sex aid can be considered. If possible, the woman places the artificial vagina between her thighs. As with the artificial penis it is essential that both partners are comfortable using the aid. The man is able to place his penis in the artificial vagina. It is advisable for him to use a condom as this allows easier cleaning of the aid after intercourse. By using the aid, the couple are able to experience the physical and emotional closeness of sexual intercourse. The ability to fantasise, or perhaps to be able to recapture memories from the past, can add to the pleasure. In the cold light of the consultation, aids do not usually appear attractive. However, if the clinician works with many people who may need aids, it is useful to be able to show them and allow them to handle it when discussing this option. Sometimes there are problems obtaining a sex aid, which may be due to physical difficulties (e.g. access to a sex shop), or there may be hesitancy about going into a shop or being seen going into a shop due to the somewhat sleazy reputation of these outlets.
Experiences with Diverse Sex Toys Among German Heterosexual Adults: Findings From a National Online Survey

We also asked participants whether they had ever used any of seven specific sex toys. If this was the case, we asked them to indicate its use within the last 12 months for solo sex and/or partnered sex, respectively. Participants were presented with a list of sex toys comprised of lubricants (e.g., water-based lubricants, silicon-based lubricants, lubricants with stimulation effects); remedies for enhancing arousal (e.g., aphrodisiacs, sex pills); toys for the stimulation of body parts (e.g., for vagina and vulva: vibrators and dildos; for penis and testicles: masturbators, cock rings, and artificial vaginas; for anus: anal dildos, butt plugs, anal beads); erotic lingerie (e.g., lingerie, latex, leather, costumes); and toys for bondage/S&M (e.g., blindfolds, cuffs, whips). The toys for the stimulation of body parts are referring to the genitals that the toy was designed/marketed for. However, those toys may be perfectly suitable across a range of body parts regardless of their intended use or the gender of its user. For each sex toy, participants were asked to indicate whether they had used the specific sex toy during masturbation/during sex with a partner in the last 12 months. Example items would be: “Sex toy for stimulation of vagina and vulva (for example vibrator, dildo etc.): I have used this toy during sex with a partner in the last 12 months”; “Sex toy for stimulation of anus (for example, anal dildo, beads, plugs, etc.): I have used this toy during masturbation in the last 12 months.” Similar items have been used by Rosenberger et al. (2012) and Satinsky et al. (2011). Items for overall and specific sex toy use had dichotomous response options (yes/no).

Semen samples were gathered from 30 Holstein Friesian breeding bulls at the Slovak Biological Services (Nitra, Slovakia). Before the collection, the artificial vagina was sterilized to avoid any secondary bacterial contamination. Strict hygiene standards were kept during the sample collection and processing. Immediately following collection, the samples were transferred into sterile pre-heated vials and transported to the laboratory within 20 minutes.

All procedures were performed using an artificial vagina, which involved a conical thin latex sleeve ending in a sterile collection tube, without external lubricant. The procedure of sampling collection was through manual stimulation, also known as manual ejaculation. Animals were previously trained. All efforts were made to minimize discomfort.
