B. vulgaris contains high amount of inorganic nitrates and Pyridine (5.6%) and 4-picolene (54.4%) are the source of red and yellow pigments known as betalains consisting of red violet betacyanins and yellow betaxanthins. Betanin is a major constituent (75–95%) of the red pigment and vulgaxanthine I and the principal pigment of the yellow betaxanthin group. Also contains amino acid or protein such as threonine, valine, cystine, methionine, isoleucine, leucine, phenylalanine, lysine, histidine, arginine, aspartic acid, serine, glutamic acid, proline, glycine, alanine, and tyrosine. Beetroot also contains isobetanin, isobetanidine, prebetanin, isoprebetanin, and vulgaxanthin II; anthocyanins and betalains.25

Amongst the betaxanthins, various types of pigments are present in plants, which include vulgaxanthin, miraxanthin, portulaxanthin and indicaxanthin (Tiwari et al. 2013). The few edible known sources of betalains are red and yellow beetroot (Beta vulgaris L.ssp. vulgaris), coloured Swiss chard (Beta vulgaris L. ssp. cicla), grain or leafy amaranth (Amaranthus sp.) and cactus fruits like those of Opuntia and Hylocereus genera (Azeredo 2009).

The content of betalains (BC) in the samples was determined according to the method described by Von Elbe et al. (2004) modified for 96 well microplate (Nunc, Roskilde, Denmark) using Multiskan GO, Thermo Fisher Scientific (Vantaa, Finland). Briefly, in the microplate well the sample was mixed with 0.05 M phosphate buffer pH 6.5 to the final volume of 250 μL. Phosphate buffer was used as the blank sample. Betacyanins and betaxanthins were measured at the wavelengths of 538 and 476 nm, respectively, and 600 nm was used for correction. Absorbances of red and yellow pigments were calculated by the following equations: a is the absorbance at 538 nm, b is the absorbance at 476 nm, c is the absorbance at 600 nm, x is the absorbance of betanin corrected for coloured impurities, y is the absorbance of vulgaxanthin-I corrected for coloured impurities and z is the absorbance of impurities. Concentration (C) of red and yellow pigments in digestates were calculated using the following equation: F is the dilution factor (25 or 12.5) and A1% is the absorbance coefficient (1120 for betanin, 750 for vulgaxanthin).

Our findings revealed that BRSD contains catechin, gentisin, 6,7-benzocoumarin, 5,7-dimethoxyflavone, and vulgaxanthin-I. Catechin, gentisin and 5,7-dimethoxyflavone are phenolic compounds, while vulgaxanthin is a natural pigment commonly found in beets. Phenolic compounds have been shown to exhibit antioxidant activity due to their hydroxyl groups that can scavenge free radicals, prevent neuronal injury and neurodegeneration [35,73]. Apart from the antioxidant activity of polyphenols, studies have shown their neuroprotective effects includes mitigation of neuronal excitotoxicity and apoptosis [74].
