In Streptozotocin-induced diabetic Wistar rats receiving a normal diet with 1% freeze-dried juice of fruits of Citrus reticulata Blanco for 10 weeks no significant impact on body weight or glycemia was observed, Serum triglycerides was decreased from 638.9 to 505.9 mg/dL (nondiabetic control: 155.7 mg/dL), low-density lipoprotein–cholesterol from 90.7 to below 67.2 mg/dL (nondiabetic group value), cholesterol from 266.4 to 208 mg/dL (nondiabetic group: 145 mg/dL). High-density lipoprotein–cholesterol was not affected.186 Aortic rings isolated from treated rodents were more respondent to endothelium-dependent acetycholine induced vasorelaxation that untreated diabetic animals with IC50 values of 63.7 and 83.3 nM, respectively. Endothelium-independent relaxation induced by sodium nitroprusside was not improved by juice supplementation suggesting a protective effect on endothelium function in diabetic rats.186 In healthy volunteers consuming fatty meals, the simultaneous intake of 1 L of red orange juice lowered 2 hours postprandial triglycerides from 173.4 to 147 mg/dL, cholesterol from 219.1 to 213.3 mg/dL, glucose from 93 to 85.7 mg/dL. This Regimen lowered arterial stiffness, neutrophil myeloperoxidase degranulation, and lowered diatolic blood pressure from 72.1 to 70.6 mmHg.187 In hypertensive patients, there is an increase of calcium concentration is vascular smooth muscle cells188 and calcium channel blockers such as nifedipine are among the most widely used drugs for treatment of hypertension. These drugs evokes reduction of vasoconstrictor tonus of the smooth muscle fibers in arterioles.189 Aurapten from Citrus spp. given at a single intravenous dose of 500 µg/kg to anesthetized normotensive rats evoked a 20 mmHg fall in mean arterial blood pressure (nifedipine 63 µg/Kg, about 30 mmHg) and an increase of heart rate from about 30 about 42 beats/min (same value for nifedipine 63 µg/kg).190

The main antioxidant genes including SOD, CAT, and GPx were analyzed and the changes in their expressions in HFF cells treated with auraptene and vitamin C as a reference antioxidant are shown in Table 3. The auraptene at the concentration of 62.5 µg/ml significantly (p < 0.05) up-regulated the expression of SOD, CAT, and GPx genes. The up-regulation of these antioxidant genes revealed the enhancement in the cellular redox state and confirmed the antioxidant potential of auraptene. The early studies reported the positive correlation of antioxidant phenolics with the cellular antioxidant enzyme activity of the treated cells (25–27). Hence in the current study, the increase in the cellular antioxidant enzyme activity could be attributed to the antioxidant activity of auraptene.
