The activity of alanyl aminopeptidase (EC 3.4.11.2) and leucyl aminopeptidase (EC 3.4.11.1) was determined spectrophotometrically as Fast Blue BB salt (4-benzoyloamino-2,5-diethoxybenzene-diazonium chloride) derivatives at 540 nm according to McDonald and Barrett (1986). The reaction was initiated by mixing 50 μl of sample and 500 μl of substrate incubation media with DMF (Serva, Germany) for 60 min incubation at 37°C, pH 6.0. Next, 500 μl of stop buffer containing Fast Blue BB Salt dissolved in 2% Tween 20 (Sigma, USA) was added, and measurements were carried out at 540 nm. For determination of alanyl aminopeptidase activity, L-alanyl-2naphtylamine in 0.1 M PBS buffer was used as a substrate. L-leucyl-2-napthylamine in 0.1 M PBS (pH 7.0) buffer was used as a substrate for determination of leucyl aminopeptidase activity.
