Sulfinpyrazone, which also possesses both uricosuric and platelet suppression properties, was assayed in plasma using detection at 254 nm and octadecylsilane columns. A mobile phase of 0.005 M tetrabutylammonium phosphate in methanol-water (56:44) flowing at 1 ml/min was used to assay the parent compound [361]. The minimum concentration was 0.2 µg/ml, and the responses are linear. Four metabolites could be resolved [362] using a mobile phase of methanol-0.02 M phosphate buffer, pH 7.0 (40:60), flowing at 1.3 ml/min. Minimum detectable limits were 30–100 ng/ml, with 85% of the parent compound recovered.

MPEG2000-PLGA18000 (50/50) was provided by A.V.T. Pharmaceutical Tech Co., Ltd (Shanghai, China). N, N-carbonyl diimidazole (CDI) and N. N-dimethylformamide (DMF) were bought from Macklin Biochemical Industry Co., Ltd (Shanghai, China). Aladdin Indust., Corp. (Shanghai, China) supplied tetrabutylammonium hydroxide and AS. AL sodium trihydrate was purchased from Shanghai Zhenzhun Biotechnology Co., Ltd (Shanghai, China). Invitrogen (Carlsbad, CA, USA) supplied 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide (MTT), RPMI-1640 medium, fetal bovine serum (FBS). HPLC methanol and absolute ethanol were of analytical grade and were obtained from Sinopharm Chemical Reagent Co., Ltd (Shanghai, China). TRACP 5 b, BGP, IL-6 and CTX-1 ELISA kits were bought from Nanjing Jiancheng Bioeng. Inst., (Nanjing, China).

The synthetic strategies used to obtain the target compounds are presented in Schemes 1–3. The O-alkylation of isovanillin (1) with bromocyclopentane was successively conducted in the presence of K2CO3 and a phase transfer catalyst tetrabutylammonium bromide (TBAB) in THF to obtain the key intermediate 3-cyclopentyloxy-4-methoxybenzaldehyde (2) that provided the core structure of phosphodiesterase-4 inhibitors37. Tetrabutylammonium bromide successively exhibited the character of phase transfer catalyst in an environmentally friendly procedure under mild conditions37.

All plasma samples were diluted five times in MPW (20%, v/v). To 50 μL of diluted plasma, ditihiothreitol was added and samples were incubated at 37 °C for 15 minutes. Then, 100 μL of 1.5 M tetrabutylammonium hydroxide was added and proteins were precipitated with acetonitrile. After a centrifugation step (10 minutes at 30,000× g, 10 °C), supernatant was diluted with MPW and transferred to sample vials.
