Vinca rosea or Catharanthus roseus, common name Madagascar periwinkle, is a fellow of Apocynaceae family, and indigenous to Madagascar and widely grown in India, Pakistan, Australia, Malaysia, and Bangladesh. In an in vivo study, aqueous extract of V. rosea was orally given to AI hyperglycemic rats for one week at a dosage of 0.4 g/kg. It was observed that the extract resulted in a noteworthy lowering in blood glucose level and the results were in par with glibenclamide, 0.01 g/kg (Raja et al., 2008). In another study, methanolic extract was fed to AI hyperglycemic rats for 14 days. It was observed that 0.5 g/kg dose was more efficient than 0.3 g/kg dose in hypoglycemic activity. This extract also ameliorated lipid profile, body weight and regeneration of β islet cells (Ahmed et al., 2010). To explore and elucidate the mechanism of action of anti-hyperglycemic activity, a study with ethanolic extract of Madagascar periwinkle was conducted. It was found that anti hyperglycemic effect was due to the complex mechanism of GLUT gene mRNA expression or amplification (Al-Shaqha et al., 2015).

Numerous naturally produced herbs that can be utilized for therapeutic purposes can be found all around us. Vinca rosea is one of them, and it's a plant that's located all over the world in tropical places. V. rosea (also known as Catharanthus roseus) is a perennial plant that may be found in tropical areas. This genus includes seven species unique to Madagascar and one member indigenous to South Asia (Johnson et al., 1963; Joshi et al., 1992; Sharma, 1998). Catharanthus roseus, Madagascar periwinkle, brilliant eyes, Cape periwinkle, cemetery plant, old maid, pink periwinkle, and rose periwinkle myrtle are some of the popular names for V. rosea. Due to anticancerous, antiviral, and diuretic properties, Catharanthus roseus is an important medicinal plant. In an experiment in which the Catharanthus plant was treated with higher carbon dioxide levels, there was an elevation in most secondary metabolites in the plant, including phenolics, alkaloids, flavonoids, and tannin (Yang et al. 2018). It was studied that, with e[CO2], Zingiber officinale showed enhancement in flavonoid and phenolic content (Ghasemzadeh et al. 2018). In Quercus ilicifolia, elevated carbon dioxide increased flavonoid and phenolic contents (Kumar et al. 2017). It was reported that enhanced content of phenols and flavonoids subsequently incremented to phenylalanine (primary metabolite), which acts as a metabolic precursor in the formation of many secondary metabolites. Other studies carried out on Labisia pumila by Ibrahim et al. (2014) also showed elevated levels of flavonoids and phenols under higher carbon dioxide levels. In addition, in the medicinal plant Pseudotsuga manziesii, there was a sudden decline in monoterpenes under e[CO2] conditions (Snow et al. 2003).

As the second most used class of anticancer drugs isolated from Catharanthus roseus G. Don. (Apocynaceae family), vinca alkaloid drugs, including Vinblastine (VBL) and Vincristine (VCR) (49) were found in an unsuccessful survey on finding oral hypoglycemic agents. The results revealed that the extract of this plant decreased white blood cells (WBC), bone density, and increased the lifespan of leukemic mice (50). In this line, VBL is now utilized in the treatment of lymphomas (51), leukemia, testicular cancer, lung cancers, breast cancer, and Kaposi’s sarcoma while VCR is used to combat leukemia (52), particularly acute lymphocytic leukemia in children (53). As other vinca drugs, Vinorelbine (VRLB) and Vindesine (VDS) are semi-synthetics, whereas Vinflunine is a new synthetic vinca alkaloid with the potential to combat the angiogenesis in cancer.
Nanomedicine-based approaches for improved delivery of phyto-therapeutics for cancer therapy

Morphine was followed by the discovery of more active entities as paclitaxel, an alkaloid extracted from the pacific yew plant (Taxus brevifolia) [3]. Paclitaxel stabilizes the microtubule polymer, by binding to β-tubulin, inhibiting the dynamic microtubule disassembly process needed for appropriate mitotic spindle formation and chromosome segregation meanwhile cell division [4]. Presently, paclitaxel is utilized in various types of cancer including ovarian, breast, lung, pancreatic, and human herpesvirus-8-induced Kaposi sarcoma and others. Also, Catharanthus roseus is the herb from which the indole alkaloids vinblastine and vincristine were isolated [5]. They are broad-spectrum anticancer drugs that attach to a site of β-tubulin other than that of the paclitaxel to prevent microtubule polymerization leading to M-phase specific cell cycle arrest.

Catharanthus (C.) roseus (L.) G. Don var. alba is a medicinally important plant associated with Apocynaceae family. Catharanthus roseus was commonly known as Periwinkle or Madagascar-periwinkle found in various tropical and sub-tropical regions [12]. Catharanthus roseus having two varieties, white-flowered “alba” and pink-flowered “rosea” [13,14]. Catharanthus roseus is an alkaloid rich plant having >400 alkaloids from which >130 are TIAs (terpenoid-indole alkaloids) [15]. Vinca alkaloids such as vincristine and vinblastine are very popular for their use in cancer treatments [14,16]. Reserpine, serpentine and ajmalicine acts as anti-neuro-inflammatory and anti-hypertensive agents [15,17]. Vindoline, vindolidine and vindolicine are important for anti-diabetic activity [18]. ZnONPs have been synthesized using C. roseus leaves [19]. However, the potential of C. roseus callus cultures for ZnONPs synthesis has not been investigated till date. Catharanthus roseus is a rich source of bioactive agents other than alkaloids, e.g. phenolics, carbohydrates, flavonoids and saponins that help in reduction and stabilization process during NPs synthesis [19,20]. Plant growth regulators (PGRs) are responsible for improved content of these bioactive agents in in-vitro cultures [21–23].
