Control of virus seed banks should include identity (genetic and immunological), virus concentration and infectious titer, virus genome integrity, expression of the therapeutic sequences, biological activity of the therapeutic sequence or the derived product (protein or RNA), sterility (bacterial, and fungal), absence of Mycoplasma and Spiroplasma (in case insect cells are used during virus seed production), absence of adventitious/contaminating viruses and RCVs (where the product is replication deficient or replication conditional), intervial homogeneity and other relevant characteristics of the virus seed bank. The complete sequence of the therapeutic and the regulatory elements and, where feasible, the complete sequence of the virus in the seed bank should be confirmed as part of the characterization (see Table 14.1).

A binary M-sequence of length 2k – 1, thus, contains all possible configurations of k consecutive elements exactly once, except the all-zero configuration. The all-zero configuration can never be part of the sequence, since the modulo 2 sum of any number of zeros is zero. These sequences are commonly called maximum length linear feedback shift register sequences or simply binary M-sequences. Note that each binary M-sequence contains a run of k – 1 zeros. In the process of video-data extraction, all the videos were separated by each signal cycle of each intersection. Each video clip contains all vehicle movements during a signal cycle. A signal cycle is a complete sequence of all signal indications. Figure A2 (Appendix, Supplementary material) illustrates the video analysis procedure in detail using the software tracker. First, real distance value was set as a calibration to obtain the proportion of conversion coordinates to pixel coordinates. The frame rate was set as five per second. Second, within each cycle’s frame, the headlamps were tracked as the feature of each vehicle that moved straight through the whole intersection. Third, the motion of headlamps of each vehicle is captured automatically by the tracker, and then trajectories consisting of particles that indicate the location of a vehicle in each frame can be extracted. In this study, only the trajectories of vehicles that go straight through the intersections were extracted to focus on the rear-end conflicts analyses.
