Careful selection of the appropriate media and incubation conditions will provide the most thorough information. Performing a justification protocol to support the chosen incubation conditions (as described earlier in this chapter) can also aid in reducing labor and the cost of consumables. Inactivators or neutralizers (e.g., penicillinase, Tween, leci-thin, and sodium pyruvate) may be needed in the media to neutralize the effects of bacteriostatic or fungistatic agents that may be picked up and transferred to the media in the process of the sampling. Some examples may be Tween and lecithin to inactivate the residues of disinfectants or preserved product aerosols, or penicillinase or cephalosporinase to inactivate/break down antibiotic powder. Sodium pyruvate can be utilized to inactivate residual vaporized hydrogen peroxide used in isolators. The need for inactivators must be researched within the context of the production operation, and the quantity of inactivator added should be in excess of what is needed but not to such excess that it becomes inhibitory to microbial growth. It is a potentially citable offense to fail to add inactivators to microbiological monitoring media where residues of a bacteriostatic agent are likely to enter the media.

The African Green Monkey Kidney cells (Vero) were procured from the National Center for Cell Sciences (NCCS), Pune, India. The cells were maintained in Dulbecco’s modified eagles medium (DMEM) supplemented with 2 mM l-glutamine and balanced salt solution (BSS) adjusted to contain 1.5 g/L Na2CO3, 0.1 mM nonessential amino acids, 1 mM sodium pyruvate, 2 mM l-glutamine, 1.5 g/L glucose, 10 mM (4-(2-hydroxyethyl)-1-piperazineethane sulfonic acid) (HEPES), and 10% fetal bovine serum (GIBCO, USA). Penicillin and streptomycin (100 IU/100 µg) were adjusted to 1 mL/L. The cells were maintained at 37 °C with 5% CO2 in a humidified CO2 incubator.

The MCF-7 breast cancer cell was obtained from the National Institute of cell science Pune in India. The MCF-7 breast cancer cell line was maintained in Dulbecco’s Modified Eagle’s Medium (DMEM) supplemented with 2 mM L-glutamine and a balanced salt solution containing 1.5 g/L Na2CO3, 1 mM sodium pyruvate, 2 mM L-glutamine, 1.5 g/L glucose, 10 mM (4-(2-hydroxyethyl)-1-piperazineethane sulfonic acid (GIBCO, USA). The concentrations of penicillin and streptomycin (100 IU/100 mL) were reduced to 1 mL/L. In a humidified CO2 incubator, the cells were kept at 37 °C with 5% CO2 (Wypij et al., 2021).
