The pharmacokinetic study was carried out in male Sprague-Dawley (SD) rats weighing 230–250 g (n = 6). Diet was prohibited for 12 h before the experiment, while water was taken freely. According to the recommended dosage of consumer available prescriptions (200 mg/times), it is converted into a rat dosage (20.57 mg/kg) [31]. Oral formulation was prepared in suspension (2 ml) form by triturating, accurately weighed amounts of powdered compounds in water with or without pre-dissolution polymers (4 mg/mL). After single oral administration to SD rats, blood was withdrawn at different times and then samples were analyzed by UPLC-MS. All experiments were executed in compliance with Animal Management Rules of the Ministry of Health of the People’s Republic of CN (document number 55, 2001) and the Animal Experiment Ethics Review of Shenyang Pharmaceutical University.

The liquisolid formulations were prepared in a three step process as follows; Firstly, a weighed amount of the Wmix was dispersed in the selected nonvolatile solvents to form a liquid medication with two different concentrations, i.e. 25 and 50% w/w. Secondly, the calculated quantity of the carrier material (MCC) was blended with liquid medication in order to achieve an even distribution of the liquid medication into the powder. Mixing took place by trituration using a mortar and a pestle. Finally, the coating material (silicon dioxide) was added and mixed thoroughly in the mortar to produce a dry looking, free flowing powder [33]. Eight liquisolid systems were prepared and their composition is presented in Table 1. For each formulation, the liquid load factor (Lf) was calculated according to Equation (2). The amounts of liquid medication (W) corresponding to 15 mg of the extracts, as well as the carrier (Q) and coating (q) materials, to be introduced in each formulation, were then determined employing Equations (3) and (4).

During the tissue dissociation, trypsin or panpain digesting technique can be used, but the enzyme concentration and its time must be noted. DNAse was required to avoid DNA wrapping over protein surface during dissociation. Cell viability is greatly influenced by trituration procedure.44 Neurons are too vulnerable and cell loss may be caused during digestion and trituration. Foaming should be avoided during trituration, for cells may be lysed at an air-liquid interface. It thereby needs very gentle trituration by a Gilson blue pipette tip.
