A number of trialkyl organotins cause the superimposition of male characteristics on normal female mud snails.21 At 4.5 to 5.5 ppb tributyltin chloride, young females show a frequency of 75% in the occurrence of male characteristics, including a 29% frequency of penis development. Older females have a reduced imposition of about 11%, and controls 5%.

Results from a series of experiments rather clearly demonstrate the following: With one major exception tin steroids showed no gross toxicity in mice when administered at 10 and 100 ppm for a 45-day period in water.Tin cholesterol compounds wherein the tin was bound through an ether linkage at 3C provided the highest tumor growth suppression index values. Several cholesterol derivatives wherein the tin was bound at 24C by an ester linkage also displayed a relatively high degree of suppression.Tin-testosterone compounds were inferior.A number of compounds caused necrotization of the tumor and what appeared to be actual killing. The superior compound in this respect was a tributyltin deoxycholate with a possible ester linkage to the tin moiety at 24C. Since this material is one of several isomers, the structure is, so far, unknown.Tributyltin chloride and tributyltin adipate were mildly tumor suppressive.Other organotins, testosterone, and cholesterol were inactive.Cholic acid, cholesterol chloride, and testeronyl-n -butylstannate accelerated tumor growth.Supporting data from an outside evaluation of several tin steroids, triphenyltin cholate and tri-n-butyltin deoxycholate showed considerable inhibition of human KB tumor and P-388 mouse leukemia in cell culture studies.Assay preferred by the National Cancer Institute based upon injection showed considerable toxicity at the higher dosage levels arising from this mode of administration, with relatively poor suppression indices. In these ongoing evaluations, best results at the time of writing were with triphenyltincholesterol ether, the same compound being one of the top three in terms of growth suppression by oral administration.
