Radix Ophiopogonis (Maidong) is the root tuber of Ophiopogon japonicus (Thunb.) Ker-Gawl. The root is sweet with a slightly bitter taste. In traditional Chinese medicine, it is used for nourishing the stomach, promoting the production of body fluid, and nourishing the lung. It is also used for nourishing the heart and the treatment of angina pectoris.

Traditional Chinese medicine (TCM), a unique medical system, has a lengthy history in China. TCM has fewer side effects and is suitable for the treatment of chronic non-serious diseases (Zheng et al. 2011). The pathogenesis of SS in TCM involves yin deficiency, dryness, heat, and blood stasis (Zhang 2011; Wang et al. 2021). Zengye decoction (ZYD), as a traditional Chinese medicine formula, is composed of Radix Scrophulariae (Chinese name Xuanshen; the dried root of Scrophularia ningpoensis Hemsl. [Scrophulariaceae]), Rehmannia glutinosa (Chinese name Shengdi; the tuber root of Rehmannia glutinosa (Gaertn.) Libosch. [Orobanchaceae]) and Radix Ophiopogonis (Chinese name Maidong; the dried tuber root of Ophiopogon japonicus (L. f.) Ker Gawl. [Asparagaceae]) with a weight ratio of 5:4:4 (Li et al. 2019). The decoction exhibits multiple pharmacological activities, such as metabolism, oxidative stress, cell ion balance, anti-inflammatory, and coagulation regulation (Yuewen et al. 2018). In clinical treatment, ZYD is used for treating constipation, SS, diabetes, pharyngitis, laryngitis, and other diseases (Minghao et al. 2016; Liu D et al. 2019; Wang M et al. 2020). Related research indicated that ZYD could lower the expression of serum TNF-α and IL-1β levels in SS model rats (Sun et al. 2015). Other research reported that ZYD protected the structure and function of submandibular glands (SMGs) in NOD mice (Li et al. 2019). Nevertheless, the mechanisms of ZYD against SS have not been elucidated.

Injection of YiQiFuMai (YQFM) powder is a modern Chinese medical preparation of natural products based on the historically proven Traditional Chinese Medicine. In China, it has been considered as a supplementary therapeutic strategy for treatment of HF with comparable ease of production and resupply, and patient compliance. YQFM, a concoction of concentrated plant extracts, consists of water-soluble compounds extracted from herbs, including the root of Panax ginseng C. A. Mey. (Araliaceae), the root of Ophiopogon japonicus (L. f) Ker-Gawl. (Liliaceae) and the fruit of Schisandra chinensis (Turcz.) Baill (Schisandraceae) (Yuan et al. 2011). YQFM's bioactive components have been identified, with ginsenosides and lignans being the most common pharmacologically active molecules (Zhou et al. 2011).

Ophiopogonis Radix, the root of Ophiopogon japonicus (Thunb.) Ker-Gawl (Liliaceae) is an evergreen perennial widely used and distributed in East Asia, especially in China. Ophiopogon japonicus is often used in compound prescriptions, such as YiQiFuMai injection, Sheng Mai Yin and Xuanmai granule. Considerable clinical evidence has showed that Sheng Mai Yin can be used for treatment of myocardial infarction, coronary heart disease and shock (Zhang et al. 2018). Extensive researches have revealed that Ophiopogon japonicus has beneficial effects on the cardiovascular diseases through various mechanisms, such as antioxidation, antiarrhythmia, improving microcirculation, etc. (Ichikawa et al. 2003; Kou et al. 2006). Phytochemical research has found that steroidal saponins are one of the main characteristic constituents of Ophiopogon japonicus. This group of compounds exhibits therapeutic effects against diabetes, chronic inflammation, oxidative stress and cardiovascular diseases (Chen et al. 2016). However, it is unknown whether saponins-rich extract of Ophiopogon japonicus (SOJ) is able to protect cardiomyocytes from DOX-induced CHF via its anti-inflammatory and antioxidative effects.
