The reference arm is more complex that typically required in a (FD)-OCT system. Ninety percent of the initial 840 nm SLD source light is directed into the reference arm via the first fiber-optic coupler. The reference light traverses polarizers to match the polarization state in the sample arm and then passes through two acousto-optic modulators (AOMs) that introduce an overall carrier frequency shift of 3 MHz. After correcting for the AOM dispersion, the beam is then input to a galvanometer-based optical delay line that can be rapidly modulated to introduce reference arm length changes to compensate for shifts in the axial position of the eye. The beam is then finally reflected off a voice coil activated mirror to provide the axial scanning before finally interfering with the sample arm light hence providing the OCT signal.

Ten microlitres of CDs (+) magnetically labelled EV fractions from two independent isolation procedures (n = 2) were fixed with 4% formaldehyde in PBS for 10 min, then they were magnetically collected and washed twice in PBS. To visualize the presence of ICAM-1 in the CDs (+) EV, samples were diluted in 1 mL PBS and incubated with ICAM-1 anti-mouse Alexa 488 (1:500) overnight at room temperature. Three hundred microlitres of magnetic beads bearing EV were then mounted onto a #1 circular cover glass. At least four images (n ≥ 4) were taken for each biological sample using a Zeiss LSM 880 confocal laser scanning microscope (Jena, Germany) on an Axiovert 200 M motorized frame. The microscope was coupled to a 30-mW, air-cooled argon-ion laser emitting at 488 nm under the control of an acousto-optic modulator (~11 μW irradiance at the sample position) for one-photon excitation. EV-depleted cell culture supernatant (sEV (-)) incubated with CDs decorated magnetic beads and served as a negative control.
