The two paragraphs translated above reflect a difference of opinion as regards the practice of gathering the original qi (yuanqi 元氣) to create the immortal embryo. Those who practise sexual union believe they can gather the original qi from the body of a woman. The authors of the Pangmenlu, and He Longxiang with them, insist that only by gathering one’s own yuanqi can one attain immortality. This is further explained in a later passage in the same text: The yuanqi within the body is the elixir of immortality. Heterodox schools and wayward ways go against reason in seeking for it outside the body; by refining the elixir and ingesting it they hasten their own death (…). Men and women come together and then extract the spoiled jing. How is this different from [what is done by] cattle and horses, dogs and pigs? They say that by eating the spoiled jing, one can become immortal; why then, do cattle and horses, dogs and pigs not ascend to the heavenly hall? This is a case of misreading the two words ‘ingest and eat’ (fushi 服食) found in alchemical books. When studying alchemical books, one must find a teacher to explain them so that one clearly understands the principles behind them.(Pangmenlu: 4b)

Yuan Point — The Yuan or source point serves as a reservoir of Qi produced in its associated organ. It is the best point to use in energy transfer or equilibration from the meridian to the organ or vice versa. The Yuan point can also tap into the original Yuan Qi from the Kidneys and is an important point to increase the supply of energy to, and to increase the metabolism of, its associated organ. The Yuan point can be acutely tender to palpation in a Yang excess pain, or deeply aching in a Yin deficiency state.

Another type of Qi is called yuan Qi, which is said to represent the energy we brought with us into this life, including ancestral energy and the energy of the soul. In systems that possess a belief in reincarnation, it represents the essence of who we were, are, and that which we will bring into our next life. Long Qi is derived from the food we eat and the air we breathe, that is, from our environmental surroundings. We also take in this type of Qi while we sleep and during sexual orgasm. For certain people, their only interaction with subtle energy will occur from elements taken from the environment, such as dreams. In other words, even without a conscious effort, humans are nurtured by subtle energy. Yuan Qi is housed in deeper meridians, called the extraordinary or curious meridians. When yuan Qi flows through the principal meridians, it is an indication that the chakras have opened to allow subtle energy to permeate the body.

Targeted NGS using the 38-gene panel (Yuanqi, Shanghai, China) was performed to determine the CALR mutation types and whether there were other nondriver genetic mutations. Genomic DNA was extracted from bone marrow samples using QIAamp DNA mini kit (Qiagen, Valencia, CA, USA). Genomic DNA quality and quantity were determined using a Nanodrop 8000 UV-Vis spectrometer (Thermo Scientific, Waltham, MA, USA), a Qubit 2.0 Fluorometer (Life Technologies Inc., Grand Island, NY, USA), and a 2200 TapeStation Instrument (Agilent Technologies, Santa Clara, CA, USA). As previously reported, this panel is designed to enrich exons of 38 genes covering 66.9 kb of the human genome (including CALR, JAK2, MPL, ASXL1, EZH2, SRSF2, IDH1, IDH2, U2AF1, TET2, STAG2, BCOR, BCORL1, DNMT3A, CBL, CEBPA, MLL, SF3B1, ATM, GATA2, NRAS, KRAS, RUNX1, PPM1D, ZRSR2, KIT, SETBP1, NF1, TP53, NPM1, PDGFRA, PHF6, PIGA, SH2B3, FLT3, STAT3, CSF3R, WT1). After the enriched exome libraries were multiplexed, the libraries were sequenced using the NovaSeq 6000 sequencing platform (Illumina, San Diego, CA, USA).

The cDNA synthesis and multiple RT–PCR were conducted by using SureFireRTTM Kit (Promega, USA) (AIBOJIN, Cat#06-104) and Leukemia Related Fusion Gene Detection Kit (YUANQI BIO), respectively. Multiple RT–PCR amplification was performed as 8 parallel multiplex reactions on 7500 Real-time PCR System (Applied Biosystems). Leukemia Related Fusion Gene Detection Kit included 41 gene fusions (BCR-ABL, SIL-TAL1, E2A-HIF, TEL-AML1, MLL-AF4, E2A-PBX1, AML1-ETO, MLL-AF9, PML-RARα, MLL-(AF6, AF10, ELL, ENL), PLZF-RARα, STAT5b-RARα, NPM-MLF1, TEL-PDGFRB, PIP1L1-PDGFRA, AML-MDS1/EVI1, AML1-MTG16, CBFβ-MYH11, DEK-CAN, TEL-ABL, ETV6-PDGFRA, NUP98-(HoxA13, HoxC11, HoxD13, HoxA9, HoxA11, PMX1), TEL-JAK2, MLL-(AF17, AF1q, AF1p, AFX, SEPT6), (NPM, FIP1L1, PRKAR1A, NUMA1)-RARα).
