One-pot synthesis and characterization of Schiff base macrocyclic complexes as a potential bioactive core – a review

Metal complexes of Schiff base macrocycles can be synthesized by the required metal salts reacting with preformed macrocyclic Schiff base ligand under specific conditions, but there are potential drawbacks in this strategy. Therefore, the best method for development of macrocyclic Schiff base complexes includes one-pot cyclization reactions where the metal ion increases the yield of the macrocyclic product [70]. The metal ions and the anions are vital as the equilibrium among the size of the anion and cation will direct the dissociation of the salt of metal in the reaction medium [79]. The metal ions act as a template for the ligand framework to take suitable geometry [80]. Through the metal template effect, synthesis of multidentate macrocyclic ligand has been archived in significant yields [8, 38]. Macrocyclization can help to make better oral absorption, pharmacological activity, cell permeability and metabolic stability for drug candidates [81].
