Biopsy dishes are prepared once it has been determined how many embryos are suitable for biopsy. Of utmost importance is embryo tracking and identification at every step of the procedure. Care must be taken to ensure that the biopsy dishes are labeled with the patient name and the number of the embryo to be biopsied. The biopsy dish is prepared with a 10 μ l drop of polyvinylpyrrolidone (circled) and a 10 μ l drop of buffered culture media for each embryo to be biopsied. At most, each dish will contain sufficient drops to perform biopsy on two embryos only. The drops are covered with pre-equilibrated paraffin oil.

Despite understanding the oil and wax composition of petrolatum, it is known that "synthetic" petrolatum cannot be created by simply combining paraffin wax and mineral oil. When this is attempted, the blend does not remain uniform and separates. Thus, it was believed at one time that a third substance was present which kept the wax-and-oil mixture stable. This material was named "protosubstance" by F. W. Breth, who claimed the discovery of this component in an unpublished study in 1925 (17).

Oil is widely used to overlay embryo culture media and can play a major role in IVF outcome. Paraffin and mineral oil are known to have different chemical properties. The polycarbon lipid tail in mineral oil contains more unsaturated bonds than paraffin oil (Vitrolife, web site), the mineral product being therefore more sensitive to photooxidation and peroxidation (Elder Key and Bw 2015). High peroxidation in mineral culture overlay is reported to be detrimental to fertilization and embryo development because of toxic contamination and/or deterioration of oil quality (Otsuki et al. 2007). The degree of peroxidation depends on heat and UV light exposure, storage conditions, manufacturer and batch number (Otsuki et al. 2007, 2009; Elder Key and Bw 2015). Some studies recommend to keep oil in a cool dark place to prevent it’s deterioration during long-term storage (Otsuki et al. 2009; Wale and Gardner 2016).

Chips were cleaned of dust and debris using laboratory tape. Microfluidic chips were then placed into 35 × 10 mm Petri dishes. Each well was loaded with kanamycin-treated OP50 in liquid NGM until a dome of the liquid droplet was visible, but no dark shadows were visible (approximately 0.5 microliters). Water was added to the Petri dish until just level with the chip surface and paraffin oil was layered over droplets and surrounding water to prevent evaporation. To conserve oil, water was used underneath paraffin oil. Before loading in individual wells, animals were picked to an unseeded plate to avoid contaminating wells with additional food. In assays with multiple genotypes, loading order always changed between trials. Animals were recorded swimming at 30 frames per second for 6 hours with a Grasshopper3 4.1MP Mono USB3 Vision camera (GS3-U3-41C6M) mounted on a Zeiss Discovery V20 microscope with a 1.25× objective providing 14.8× magnification. For image capture, we used FlyCap2 version 2.12.3.2 or SpinView version 1.13.0.33. Image resolution was 2048 × 1600 pixels, 270 × 270 pixels per chamber with approximately 1,800 pixels per animal. After the morphological examination, the rest of the rats in the three groups were re-anesthetized to assess the erection elicited by DNP stimulation. After making a 1-cm midline incision in the penile root region, the penis was freed from the skin, and the right branch of the DNP was carefully isolated. After that, a 24-gauge needle mounted on a PE 50 catheter, which was filled with heparin saline (250 U/mL), was inserted into the lateral surface of the corpus cavernosum and connected with a pressure transducer. The isolated DNP was stimulated with a bipolar hook electrode using continuous square-wave pulses (1 mA, 40 Hz; pulse width, 0.1 ms; duration, 60∼80 s).19 Paraffin oil was applied to protect the nerve from dryness and insulate the electrode from surrounding tissues. The intracavernous pressure (ICP) signal converted by the transducer was processed and recorded by BL-420F biological signal converter system (Chengdu TME Technology Co., Ltd., China). Nerve stimulation was repeated in each rat three times. The maximal increase of ICP (ΔICP) and the average latency from the start of the stimulus to the beginning of the ICP rise were used for statistical analysis.19,20
